Exclusive: U.N. rights forum poised to condemn Syria (Reuters)

GENEVA (Reuters) ? The top United Nations human rights forum is expected to condemn Syria for crimes against humanity at an emergency session on Friday, European and Arab diplomats said.

The move is also designed to put pressure on China and Russia to take a stronger stand against the government of President Bashar al-Assad, they said.

More than 20 member states back the U.N. Human Rights Council holding a special session, which is to be announced on Wednesday, they added.

The 47-member forum's third session on Syria in eight months is being convened days after a U.N. commission of inquiry said Syrian security forces had committed murder, torture and rape during their crackdown on pro-democracy protests.

"This is very much being led by the Arab group. Some Arab ambassadors are at least as concerned as the European Union and United States and possibly more," Britain's ambassador Peter Gooderham told Reuters.

"There is no question that the resolution will be very hard-hitting at the Council's session on Friday," he said. "It is all intended to build up the maximum pressure that the Human Rights Council can apply."

An Arab diplomat in Geneva, who declined to be identified, told Reuters: "Arab support is there, the three Gulf Cooperation Council countries who are Council members -- Qatar, Kuwait and Saudi Arabia -- and Jordan. I'm sure Libya will be with us too.

"It is meant to be a tool for later action in New York," he said, referring to the General Assembly and Security Council.

On Sunday, the Arab League imposed sanctions on Damascus over its crackdown, in which more than 3,500 people have been killed since March, including 256 children, according to the United Nations. The EU weighed in one day later, further tightening the financial screws on Damascus.

Turkish Foreign Minister Ahmet Davutoglu said on Tuesday Turkey would announce its sanctions soon.

"GROSS VIOLATIONS"

An EU draft resolution to be presented to the U.N. rights forum for adoption, obtained by Reuters, strongly condemns "continued widespread, systematic and gross violations of human rights and fundamental freedoms by the Syrian authorities".

These included executions, killing and persecution of protesters, activists and journalists, as well as arbitrary detentions, enforced disappearances and torture.

"The resolution will certainly be adopted, no question, it has wide support," the Arab diplomat said.

The draft text recommends that the General Assembly consider the commission of inquiry's report and refer the report to the Security Council "for its consideration and appropriate action".

The Security Council has the power to refer a country to the prosecutor of the International Criminal Court (ICC).

"This is probably the most controversial element in the resolution," Gooderham said.

Russia and China, which both have oil concessions in Syria, teamed up last month to veto a Western-backed Security Council resolution condemning Assad's government for violence.

The British envoy, asked about chances of winning support from China and Russia for the resolution, said: "Naturally we hope they will support this and not call a vote on the resolution as they did in August where they found themselves in a very small minority of only four states," he said.

"We hope that they will look at the merits. The seriousness of the evidence is incontrovertible," he said.

The U.N. commission of inquiry, which interviewed 223 victims and witnesses including defectors from the Syrian army, catalogued executions, torture, brutal rapes and abductions.

"They didn't even have access to Syria. Imagine what is happening really inside the country," the Arab diplomat said.

"It is close to the Iraq of Saddam Hussein, it is not any different in terms of human rights," he said.

Oil falls to near $99 after US supplies increase (AP)

SINGAPORE ? Oil prices fell to near $99 a barrel Wednesday in Asia after a report showed U.S. crude inventories rose more than expected, suggesting demand is tepid.

Benchmark crude for January delivery was down 65 cents to $99.14 a barrel at late afternoon Singapore time in electronic trading on the New York Mercantile Exchange. The contract rose $1.58 to settle at $99.79 on Tuesday.

In London, Brent crude was down 11 cents at $110.71 on the ICE futures exchange.

The American Petroleum Institute said late Tuesday that crude inventories rose 3.4 million barrels last week while analysts surveyed by Platts, the energy information arm of McGraw-Hill Cos., had predicted an increase of 1 million barrels.

Inventories of gasoline fell 173,000 barrels last week while distillates gained 1.4 million barrels, the API said.

The Energy Department's Energy Information Administration reports its weekly supply data later Wednesday.

Crude has jumped from $75 early last month amid growing expectations the U.S economy will avoid a recession. However, some analysts, such as Richard Soultanian of NUS Consulting, expect Europe's debt crisis will help trigger a recession on the continent during the next few months, which would undermine global crude demand.

"Energy market fundamentals remain weak," Soultanian said. "Europe's worsening economic troubles will negatively impact an already weak U.S. economy as well as a Chinese economy fighting a property bubble."

Analysts are also eyeing the gradual return of Libyan oil production to global supplies. Morgan Stanley said Brent prices could fall as low as $85 in the first half of next year.

"The significant supply increases and slowing demand could all coalesce around the second quarter," Morgan Stanley said in a report. "Given this backdrop, if OPEC were to leave production at current levels, inventories would balloon through the first half."

In other Nymex trading, natural gas fell 1.6 cents at $3.62 per 1,000 cubic feet. Heating oil slid 1.5 cents to $3.02 a gallon and gasoline futures dropped 0.3 cents to $2.54 a gallon.

Bush embryonic stem cell lines different from newly derived cell lines

Established human embryonic cell lines, including those approved for federal research funding under former President George W. Bush, are different than newly derived human embryonic stem cell lines, according to a study by UCLA stem cell researchers.

The finding, by scientists with the Eli and Edythe Broad Center of Regenerative Medicine and Stem Cell Research at UCLA, points to the importance of continuing to derive new stem cell lines so researchers can better understand pluripotency, the ability of these cells to make every cell in the human body, said study senior author Amander Clark, an assistant professor of molecular, cell and developmental biology in Life Sciences.

"It is critical to find out the characteristics that result in the highest quality pluripotent stem cell lines that we can make," Clark said. "It is possible that we have not set the bar high enough yet for embryonic stem cells or induced pluripotent stem cells. We now know that established lines are different from newly derived lines and now we have to find out how important that is."

The study appears Nov. 30, 2011 in the early online edition of the peer-reviewed journal Human Molecular Genetics.

The study looked at the first six human embryonic stem cell lines developed by Clark's research team at UCLA from 2009 to 2011, which have since been accepted by the National Institute of Health's embryonic stem cell registry, founded by executive order in March 2009. Acceptance into the registry allows the UCLA lines to be used in federally funded research projects.

In her study, Clark decided to examine X chromosome inactivation and the mechanisms by which female stem cells turn off one X chromosome during development because it is a large physical marker that is easy to visualize in individual cells. Clark wanted to compare this specific molecular signature in established embryonic stem cell lines versus what occurs during the derivation of new embryonic stem cell lines from human blastocysts.

The established lines examined in the study were from a group of stem cell lines derived prior to 2001. The field has known for many years that the majority of established lines, Clark said, had already undergone X chromosome inactivation, and her work confirmed this finding. However, with the progression of time, Clark found that the molecular signature no longer reflected the normal process of X chromosome inactivation.

The X chromosome normally is inactivated by non-coding RNA and a special form of chromatin in female cells. In abnormal states, such as those found in the older, established human embryonic stem cells, the X chromosome is inactive, but this process is not regulated by the non-coding RNA and the chromatin is different.

"The classic signature is gone, so something else is regulating X chromosome inactivation in the established cell lines," Clark said. "It will be important not only to find out what that is, but also to discover what else is changing in the nucleus that we cannot see regardless of whether the cell line is male or female."

The new cell lines generated by Clark's research team were derived from human embryos that were donated to the Broad Stem Cell Research Center by couples who had previously undergone in vitro fertilization to overcome infertility. The couples no longer planned to store or use their frozen embryos for reproductive purposes and had declined to donate the embryos to others for reproductive use.

The human embryos were transferred from the fertility clinic to the derivation lab at UCLA in frozen vials. They were then thawed by Clark's research team, and at six to seven days of development the embryos, or blastocysts, contained a cluster of cells called the inner cell mass. The inner cell mass is the source of new embryonic stem cell lines.

Clark's lab examined the human embryonic stem cell lines three to four weeks after growth from the inner cell mass and found that both X chromosomes were still active in many cells, making them more like the cells from the original inner cell mass.

Slowly, with time in culture and cryo-preservation ? how the lines are ultimately stored - one X chromosome is inactivated and the cell lines become identical to the older, established lines, including abnormal X chromosome inactivation, Clark said.

The question, Clark said, is whether the first cells to grow out from the inner cell mass are of a higher quality, and therefore the ones researchers should be aspiring to use for research and potentially therapeutically.

"It may prove to be important to stabilize these cells at that very young state, one that's closest in identity to the inner cell mass," Clark said. "And then we can ask whether these cells give the best quality when differentiated into clinical cell types."

Keeping both X chromosomes active will also be important in modeling diseases such as Rett syndrome.

Going forward, Clark will study human embryonic stem cells in three states, lines in which the X chromosome is inactivated by normal means, lines in which the chromosome is inactivated abnormally and lines in which both X chromosomes remain active. Clark will seek to understand the differentiation potential of each of the three states.

"Our data highlights the importance of maintaining human embryonic stem cell derivation efforts. Gold standard human embryonic stem cell lines should be the benchmark for all human pluripotent stem cell research," the study states. "Developing new experimental approaches aimed at sustaining human pluripotent nuclei in an epigenetic state closer in identity to the day six or seven day human blastocyst is to work towards a more robust gold standard."

This work presents the technical possibility to carry out within a day after the release of new economic data for the Spanish quarterly accounting, quarterly forecasts for GDP economic growth (Gross Domestic Product) for all of the autonomous regions. This forecast has proven to be reliable and consistent with GDP growth in the Spanish economy, according to Antoni Espasa, Full Professor of Econometrics in the UC3M Statistics Department, one of the authors of this study, in which Angel Cuevas and Enrique M. Quilis, from the Ministry of Tourism, Industry and Trade, and the Ministry of Economy and Finance, respectively, have also participated. The study, entitled Combining Benchmarking and Chain-Linking for Short-Term Regional Forecasting, was presented last year at the DIW Econometric Workshop in Berlin (Germany), and this year at the International Symposium on Forecasting, in Prague (The Czech Republic) and the IWH-CIREQ Macroeconometric Workshop, in Halle (Germany).

This line of research could have applications in the current economic context. In this work, for example, the methodology described is applied to give a quarterly profile of economic growth in each autonomous region for the last 16 years, and the forecasts for the rest of 2011. "This allows us to see the differences in the economic cycles particularly in the latest recession and the budding recovery which began in 2010," Antoni Espasa pointed out. "The application," he added, "gives forecasts for growth for all of the autonomous regions that should relate them with budget tightening plans and public deficit objectives." In the current economic context which is marked by uncertainty and volatility, a tool such as the one presented in this work contributes to clearly improving analysis of the current economic situation and corresponding decision making, according to Professor Espasi.

Forecasting economic times

The data necessary to carry out this type of predictions are those related with the Los Contabilidad Nacional Trimestral de Espaa (CNTR) (National Accounts Data), and the Contabilidad Anual Regional de Espaa (Regional Accounts Data), as well as the monthly and quarterly data on main macroeconomic indicators from each Autonomous Region. Next, econometric techniques of time series models are applied to obtain the forecast, seasonally adjusted and interpolation with reference index. With that, a new instrument is obtained for short-term monitoring which allows analysts to quantify the degree of synchronization among regional economic cycles.

This research is within the framework of the commitment made by the UC3M Instituto Flores de Lemus to analysis and diagnosis of the economic reality. The researchers of this center, lead by Professor Espasa, have a great deal of experience, dating back more than 17 years, in monitoring, forecasting and diagnosis of the real economy, with its monthly bilingual publication of Boletn de Inflacin y Anlisis Macroeconmico.

MTV's fiercest bad girl, Kristin Cavallari, and her football-player beau, Jay Cutler, are engaged again.

"The Hills" alum confirmed the news Wednesday on Twitter. "This time it's official..Jay and I are engaged again," she wrote.

News of a possible reconciliation between the onetime "Dancing With The Stars" competitor and the Chicago Bears quarterback first hit the rumor mill earlier this month when Life & Style reported that the twosome had decided to give it another go after ending their first engagement over the summer.

However, at the time, the couple denied they had set the wedding wheels back in motion. Us Weekly reports that Cavallari was spotted Tuesday in Los Angeles wearing the 5.2-carat engagement ring he had given her for their first engagement.

The couple announced their original engagement in April when he popped the question during a vacation in Mexico. However, the first engagement ended only months later in July. The two had reportedly been deep in wedding planning when they called it off.

"She got dumped. She's absolutely devastated. She can't believe this is happening," a source said at the time. "She was planning her dream wedding, the date was set, the location — everything was set. She's in shock that the dream wedding she was planning is going to end this way."

Back before she decided to settle down with Cutler, Cavallari had been one of reality TV's greatest players. During her time on "Laguna Beach" and "The Hills," she spent her time with guys like Justin Bobby, Brody Jenner and Stephen Colletti.

Can You Afford a Break?

The first question to ask yourself when considering your new job's start date is how long you can afford to be out of a job. In some cases, your new employer may want you on the job as soon as possible, and if you can't afford to take time off in between jobs, even for a few days or a long weekend, then the decision is already made for you. If you can afford a few days away from work, have your own emergency fund, or just need some time to recharge, you may want to consider a couple of days in between jobs to help you get in the right mindset to start a new gig. Here are some basics you should consider.

  • Can you survive missing a paycheck? Make sure you can afford to take a few days off to relax before you make the decision.
  • When does your new employer want you to start? You want to make a good impression on your new employer, and while most won't mind if you want a few days off between jobs, if your new employer wants you to show up the Monday after your last Friday at your old job, you may have an uphill battle trying to squeeze in a few days off in between to relax. Make sure your new employer is okay with you delaying your first day, or schedule your last day at the current employer so you can get a few days off between jobs.
  • When do your benefits expire? For many of us, leaving a job means losing our health insurance and other benefits. Do your benefits expire the day you leave the company, or at the end of the month? When do the benefits at the new job kick in? Do you anticipate any medical needs if you have any periods without insurance? Consider your 401(k), and if you need to roll it over or make any changes to it, or if there's any paperwork you have to complete before you can start a new job. Think these things over carefully before deciding to take a few days off. If your benefits expire at the end of the month or your new benefits won't take effect for a month after you start anyway, there's no harm in taking a couple of days between jobs.
  • Do you need time to recharge? If you're leaving your current job because you're burnt out and just need time to get away from what you're doing, you should definitely consider a little time between jobs. You don't want to start the new job carrying all of the emotional baggage from your last job?you want to go in with fresh eyes, eager to learn new things and meet new people. You shouldn't be world-weary on your first day at a new gig.
  • Do you have questions for your new boss that you want answered before you get there? Are there any lingering questions you might have about the position that you want answered before you show up? You may want to take a day or so to get your thoughts in order and ring them with questions or thoughts so you can hit the ground running when you arrive on your first day.

Once you have the basics out of the way, you can address some more personal questions and considerations that might make you want to take a few days between jobs.

Make sure you talk with your future manager about what your company's equipment policies are and what your company will provide you with, and then go shopping for the rest.
